Description:
The 3×Flag Peptide is a synthetic peptide of 23 amino acid residue. The Asp-Tyr-Lys-Xaa-Xaa-Asp motif is repeated three times in the peptide. Eight amino acids at the C-terminus make up the classic Flag sequence (Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys).
Benefits:
After competitive elution of the Flag tag fusion expression protein from this product, the eluted sample does not contain the light and heavy chains of the traditional Anti-Flag tag antibody, effectively solving the interference issue of light and heavy chains during Western detection after immunoprecipitation. In conventional immunoprecipitation, the Flag tag fusion expression protein is usually released into the solution by boiling with the addition of loading buffer, but at this time, the light and heavy chains of the Anti-Flag tag antibody also enter the solution and are likely to interfere with subsequent Western detection. The most important thing about peptide elution is to maintain the activity of the elution product.
Recommended working concentration is 100-400μg/ml for elute 3×Flag fusion proteins from the Flag tag affinity gel(DYKDDDDK tag Nanoselector Agarose, DYKDDDDK tag Nanoselector Magnetic beads or other Anti-Flag affinity gel).
Dilution factors are presented in the form of a range because the optimal dilution is a function of many factors, such as antibody density, permeability, etc. The actual dilution used must be determined empirically.
