Description:
DogCatcher Magnetic beads have been specifically designed to bind DogTag-fusion proteins. DogCatcher Magnetic beads are based on Dogcatcher covalently coupled to the surface of magnetic beads, and they are used to detect and isolate DogTag-fusion proteins efficiently.
Ligand: DogCatcher
Bead size: ~ 40 µm
Reactivity: Highly selective for DogTag-fusion proteins.
Binding Capacity: High binding capacity, 10 µL slurry bind about 10-20µg of DogTag-fusion protein.
Storage: Shipped at ambient temperature. Upon receipt store at 4°C. Stable for 1 year. Do not freeze.
Storage Buffer: Supplied in PBS Buffer (pH 7.2), 0.1% Tween 20.
Background:
The DogCatcher-DogTag system was developed seven years ago as a method for protein ligation.
It is based on a modified domain from a S. pneumoniae surface protein (RrgA). Upon recognition, the two form a covalent isopeptide bond between the side chains of a lysine in DogCatcher and an Asparagine in DogTag. This technology has been used, among other applications, to create covalently stabilized multi-protein complexes, for modular vaccine production, and to label proteins. The DogTag system is versatile as the tag is a short, unfolded peptide that can be genetically fused to exposed positions in target proteins; similarly, DogCatcher can be fused to reporter proteins such as GFP, and to epitope or purification tags. DogTag/DogCatcher can be used for rapid loop-mediated labeling of ion channels in live mammalian cells.
